Project

Telomere dynamics in C.elegans

Telomere is composed of repetitive DNA and its sequence-specific DNA-binding proteins. The telomere double-stranded DNA-binding proteins are evolutionally conserved and characterized from yeast to human model systems. However, there is no apparent homologs of such telomere-binding proteins in nematode C.elegans, suggesting hitherto unknow proteins might engage in telomere protection in C.elegans. We have recently identified the long-awaited telomere double-strand DNA-binding proteins in C.elegans, which we named DTN-1/2 (double-strand telomeric DNA-binding protein-1/2). Preliminarily, we have found that DTN-1/2 have redundant roles in the suppression of the DNA damage response (DDR), while they play opposing roles in the regulation of telomere length (i.e. DTN-1 is a negative regulator of telomere length while DTN-2 is a positive regulator). This training program is aimed at developing the relevant skills in analyzing the molecular mechanism of how DTN-1/2 ensure their functions as the protector of telomere and the regulator of telomere length homeostasis. The postdoctoral fellow will study the telomere dynamics in nematode C.elegans. Analysis will include C.elegans genetics, EMS screening, CRISPR/Cas9, Immunohistochemistry, Immunoprecipitation, and biochemistry. Candidates must have basic knowledge and experience in C.elegans model system.
