Project Summary
This 12-month Laboratory Research Scientist position will support research on epigenetic mechanisms important for cancer development within the Scaffidi Laboratory.
Key Responsibilities
• To ensure continuation of an ongoing project involving analysis of mouse embryos depleted of the linker histone H1.0. We have shown that H1.0 is an important inhibitor of cancer progression in numerous cancer type (Torres at al. Science 2016, Morales et al. Nature Comms 2020). We have recently found that H1.0-deficient mice show a strong embryonic lethal phenotype associated with cell hyperproliferation and we are characterizing the molecular basis of this effect. The project involves analysis of embryos by immunofluorescence microscopy using established protocols, and molecular characterisation of embryonic stem cells that enable temporal control of H1.0 depletion.

Key Experiences and Competencies

Essential
• BSc in Biochemistry, Cell Biology or related disciplines, or at least 5yrs of relevant laboratory experience.
• Experience in basic laboratory techniques such as cell culture, PCR, genomic DNA extraction, qRT-PCR, microscopy
Desirable:
• Highly desirable: Experience with animal work (mice)
• Experience with confocal microscopy
• PhD in a relevant discipline
• Experience working with GM mice
• Experience with the use of databases
• Experience with animal tissue handling (collection, dissociation, preparation of FFPE sections or cryosections)
• Holder of UK Animal scientific procedures personal license
• Experience with mouse embryonic stem cells
The post holder will be trained in all specific techniques by a Principal Laboratory Scientist and will receive support by the rest of the lab and the Crick core facilities.
